931 Siglec-E expression on innate immune cells is enhanced in chronic skin inflammation

Human sialic acid-binding immunoglobulin-like lectin-9 (Siglec-9) and mouse homologue Siglec-E are members of Siglec family, which expresses on various immune cells works mainly as an inhibitory receptor. Although Siglec-9 have been recently indicated about their functions in some inflammatory diseases, there is no report skin diseases. To study a relation between we used models acute allergic dermatitis (AAD), irritant contact (ICD), chronic (CAD). As AAD model, ears were elicited with 2,4,6-trinitrochrolobenzene (TNCB) after sensitization. CAD TNCB was applicated repeatedly onto elicitation. ICD stimulated by phorbol myristate acetate. We first compared gene expression cytokines via quantitative polymerase chain reaction (qPCR). qPCR showed that mRNA expressed model fiftyfold higher than (p < 0.05) did not express similar tendency cytokines. next checked whether keratinocytes or immunocytochemistry (ICC) primary keratinocyte cultures, isolated from newborn mice. ICC cultures Siglec-E. finally examined tissue immunohistochemistry (IHC). IHC ear samples more highly macrophages neutrophils T increased AAD. Taken together, our indicates cells, innate AAD, enhanced inflammation inflammation.
